Evaluation of an indirect ELISA using recombinant granule antigen Gra7 for serodiagnosis of Toxoplasma gondii infection in cats.

The precise detection of Toxoplasma gondii infection in cats has important public health significance. In the present study, recombinant granule antigen protein GRA7 was evaluated as a potential diagnostic marker for T. gondii infection in cats by an indirect enzyme-linked immunosorbent assay (ELISA), using the classified serum samples from cats, by immunofluorescence assay (IFA) and by modified agglutination test (MAT). There was a perfect agreement (97.2%) between GRA7-ELISA and MAT/IFA (Kappa = 0.92; 95% confidence interval [CI], 0.85 to 0.99), and GRA7-ELISA had a sensitivity of 94.9% and a specificity of 97.9%. No significant difference (P > 0.05) was observed between the detection results by GRA7- and Toxoplasma lysate antigen-based ELISA. Receiver operating characteristic analysis showed a relative sensitivity and specificity of 89.7 and 92.5% at the cut-off value of 0.1 for GRA7-ELISA. These data demonstrate that GRA7 is a promising serodiagnostic marker for T. gondii infection in cats.

Genetic characterization of Toxoplasma gondii isolates and toxoplasmosis seroprevalence in stray cats of İzmir, Turkey.

Currently, some Toxoplasma gondii genotypes are being associated with serious clinical presentations. A recent report showing the Africa 1 genotype in two local congenital toxoplasmosis cases acquired in Turkey formed the basis of this study because atypical Africa 1 genotype is most frequently detected in animals and patients from sub-Saharan Africa. Since stray cats are considered as the linkage between wild life and urban life in T. gondii transmission, the present study aimed to isolate and characterize T. gondii strains circulating in stray cats of İzmir (Western Turkey). A secondary objective was to determine toxoplasmosis seroprevalence in this cat population. Tissues obtained from 100 deceased stray cats were bioassayed and isolated strains were genotyped using 15 microsatellite markers. In addition, toxoplasmosis seroprevalence was analyzed in 1121 cat sera collected from several large veterinary clinics in İzmir. Among the 22 isolates, 19 were Type II (86.3%), two were Type III (9%) and one was Africa 1 genotype (4.5%). The overall seropositivity rates in cats were 42-48% and 33.4-34.4% according to IFA and ELISA, respectively. Seroprevalence in deceased cats was significantly higher than in healthy cats (P = 0.0033). Finding both the major clonal Type II lineage together with the Type III lineage also found in Middle East, and an atypical genotype, Africa 1 appears consistent with the specific geographic location of Turkey between three continents and raises the possibility of transportation of these strains between continents through trade routes or long distance migratory birds. In addition, the first large study of toxoplasma seroprevalence in a stray cat population was also reported. The relatively high seropositivity rates and the variety of T. gondii genotypes confirm the local stray cat population as a risk factor for human toxoplasmosis in İzmir.

Prevalence of IgG antibodies to Encephalitozoon cuniculi and Toxoplasma gondii in cats with and without chronic kidney disease from Virginia.

Kidney disease is a common and serious condition in domestic cats. There are numerous causes of kidney disease including parasitic infection. Encephalitozoon cuniculi is a microsporidian parasite that develops in the kidneys of rabbits and causes chronic renal disease. Little has been reported concerning E. cuniculi in cats and no serological studies on this parasite in cats have been conducted in the United States to date. The present study explored the possibility that E. cuniculi is an unrecognized contributor to the high prevalence of kidney disease observed in cats. A serological survey was conducted to determine the prevalence of IgG antibodies to spores of E. cuniculi in cats with and without a diagnosis of chronic kidney disease (CKD) according to the International Renal Interest Society (IRIS) staging system. Likewise, samples were examined for IgG antibodies to Toxoplasma gondii, a common well studied protozoan of cats. Plasma and sera were obtained from 232 feline patients at the Virginia-Maryland Regional College of Veterinary Medicine teaching hospital. With the investigators blinded to the renal status of test subjects, samples were screened via indirect immunofluorescent antibody assay (IFA). Thirty-six of the 232 cats met the IRIS staging system criteria for CKD. Antibodies to E. cuniculi were found in 15 of the 232 samples, which included 4 of the 36 cats with CKD. Sera from cats serologically positive to E. cuniculi did not react to spores of E. intestinalis or E. hellem when examined in the IFA. Antibodies to T. gondii were found in 63 of the 232 samples, which included 10 of the 36 cats with CKD. The prevalence of antibodies in cats with CKD to either protozoan was not significantly different (P>0.05) from the cats without CKD in the study. Collectively the results do not support the hypothesis that either E. cuniculi or T. gondii play a significant etiologic role in the occurrence or progression of CKD in cats.

Presence of domesticated cats and visual impairment associated to Toxoplasma gondii serum positive children at an elementary school in Jataizinho, state of Paraná, Brazil.

Toxoplasmosis possess worldwide geographic distribution and high serological prevalence in human beings and animals. Acquired way is, in the majority of the times, light or asymptomatic however, in approximately 15% of the cases can be developed the ocular form. The aim of this study is to research the occurrence of IgG anti- Toxoplasma gondii antibodies in children at an elementary school in Jataizinho (PR) and relate some factors with the epidemiology of the toxoplasmosis. A total of 276 samples of blood by using indirect immunofluorescence (IFA) where 128 (46.4%) were detected positives. The analysis of the variables indicated that the presence of domestic cats in the household was an important factor associated to the infection by T. gondii (OR= 3.45; 1.61- 7.45) as well as to the children who described any kind of visual impairment (OR= 3.19; 1.11 - 9.35).

CpG-oligodeoxynucleotides enhance porcine immunity to Toxoplasma gondii.

Protection against a challenge infection with Toxoplasma gondii VEG strain oocysts was examined in pigs after vaccination with T. gondii RH strain tachyzoites with or without a porcine specific synthetic oligodeoxynucleotides (ODN) containing immunostimulatory CpG motifs. Six groups of pigs were immunized with incomplete Freund's adjuvant (IFA) and either vehicle, tachyzoites alone or in combination with three different doses of CpG ODN or with CpG ODN alone. Protection from challenge was significantly (P < 0.05) improved in pigs vaccinated using CpG ODN as an adjuvant with tachyzoites compared to all other groups. The CpG ODN tachyzoite-immunized pigs also had higher serum parasite specific IgG antibody, no clinical signs of disease, and 52% had no demonstrable tissue cysts after the challenge infection. These data indicate that CpG ODN is a potential safe and effective adjuvant for the T. gondii RH strain vaccine in pigs.

Toxoplasmosis in last four years in Agean region, Turkey.

Anti-Toxoplasma antibodies were determined with IFA and ELISA tests in 9410 patients who were different age groups, attended Department of Parasitology between 1991-1995. Anti-Toxoplasma IgG antibodies were found positive in 4651 (49.4%) of these patients. 2287 (21.4%) patients were pregnant women and the positivity was 55% of them. According to history of these patients, seropositivity was found 50% in women having spontaneous abortion, 52% in women having stillbirth, in 55% women having abnormal fetal births. These patients and their culinary habits, the presence of cats and relationship with other clinical symptoms were also evaluated.

The prevalence of Toxoplasma gondii antibodies among cats from Mahalla El-Kobra, Gharbia Governorate.

Toxoplasma gondii antibodies were detected using indirect haemaggulination (IHA) and indirect fluorescent antibody (IFA) tests. Positive reactions were detected among 18.5% and 12.5% of stray (92) and owned cats (32) using IHA-test respectively. With IFA-test antibodies were 20.7% and 15.6% respectively.

Toxoplasma gondii infection of cats in Beirut, Lebanon.

The prevalence of serum antibodies to Toxoplasma gondii was determined by the indirect fluorescent antibody (IFA) test in cats of Beirut, Lebanon from September 1980 to July 1983 (35 months). Testing revealed that 253/324 (78.1%) cats had antibodies in the IFA test. While the prevalence of Toxoplasma antibodies in stray cats was considerably greater than in owned cats at the beginning of the study, this difference diminished over time, and prevalence was similar by the end of the study. Faecal shedding of Toxoplasma-like oocysts was found in 31/313 (9.9%) of cats, while 38/313 (12.1%) shed other Isospora spp. oocysts. The occurrence of dual infections was statistically significant.
